dc.description.abstract | A study was conducted on the circulation of potentially diarrheagenic Escherichia coli in two groups of children, both under the age of seven. The first group (548 children) suffered from mild diarrhea and attended the Xipamanine Health Center of Maputo, in Mozambique. The second group (380 children) included randomly chosen, asymptomatic, children from the same population. A total of 503 E. coli strains were isolated from the two groups of children (n = 375 and 128, respectively). All E. coli strains were genotypically and phenotypically screened. The presence of virulence-associated genes was assessed by a set of multiplex PCR spe- cific for st and lt genes of enterotoxic Escherichia coli (ETEC), eae and bfpA genes of enteropathogenic E. coli (EPEC), stx 1 and stx 2 of enterohemorrhagic E. coli (EHEC), ial of enteroinvasive E. coli (EIEC) and the species-specific gene uidA. Adhesion and citotox- icity of isolated E. coli were evaluated in vitro on different cell cultures. A total of 37 isolates harbored virulence-associated genes: 18 were classified as ETEC, (15 from symptomatic, and three from asymptomatic children), 16 as EPEC (respectively, 13 and 3) and three EIEC in the symptomatic group. No stx 1 or stx 2 genes, associated with enterohemorrhagic E. coli were found. On the basis of the adhesion pattern on HeLa cells, 167 E. coli were classified as diffusely adhering, (125 in patients and 42 in controls) and 67 as enteroaggregative, (50 and 17, respectively). To the best of our knowledge, this is the first report in the literature on the circulation of potentially diarrheagenic E. coli in Mozambique. | en_US |

O primeiro grupo (548 crianças) sofria de diarreia ligeira e frequentava o Centro de Saúde Xipamanine de Maputo, em Moçambique. O segundo grupo (380 crianças) incluiu crianças assintomáticas escolhidas aleatoriamente da mesma população. A um total de 503 cepas de E. coli foram isoladas dos dois grupos de crianças (n = 375 e 128, respectivamente). Todas as cepas de E. coli foram rastreados genotipicamente e fenotipicamente. A presença de genes associados à virulência foi avaliada por um conjunto de amostras de PCR multiplex. específico para genes st e lt de Escherichia coli enterotóxica (ETEC), genes eae e bfpA de E. coli enteropatogênica (EPEC), stx 1 e stx 2 de E. coli enterohemorrágica (EHEC), ial de E. coli enteroinvasora (EIEC) e o gene específico da espécie uidA. Adesão e citotox- A toxicidade de E. coli isolada foi avaliada in vitro em diferentes culturas celulares. Um total de 37 isolados continham genes associados à virulência: 18 foram classificados como ETEC, (15 de crianças sintomáticas e três de assintomáticas), 16 como EPEC (respectivamente, 13 e 3) e três EIEC no grupo sintomático. Não foram encontrados genes stx 1 ou stx 2 associados a E. coli enterohemorrágica. Com base em o padrão de adesão nas células HeLa, 167 E. coli foram classificadas como de adesão difusa (125 em pacientes e 42 em controles) e 67 como enteroagregativo, (50 e 17, respectivamente). Até onde sabemos, este é o primeiro relato na literatura sobre a circulação de E. coli potencialmente diarreiogénica em Moçambique.(TRADUÇÃO NOSSA) | en_US |
